How are readings programmed to allow customers to understand where buses are going?

It could help for those who take the bus and use the Airtrain.

But who does that?

Why do so many bus routes say the street they travel on in only one direction? The S55, S74 and S79 SBS all say "via x" in only one direction of travel. Why not the other way?

The same could be asked for why the Q84 says via 120 Av in one direction, and the Q26 via 46 Av. Your guess is as good as mine

Usually it's because the street is further up in the one direction.

If you're coming from the east, the Q88 via 73rd doesn't make any sense, because the Q88 goes via 73rd first. On the other hand, knowing that the Q88 goes via Horace Harding is way more important coming from the east than it is on the west, where you would likely catch the bus on Horace Harding anyways.
